Den Hollander was born in Zambia. Her father was a miner from Northern Ireland and her mother came from Liverpool, England. Neither had completed school. She is the eldest of four children who grew up in a small South African gold mining town, and the first member of her family to go to university.

Her PhD thesis was in the scientific area and subsequently her academic career focussed on cellular biology, biochemistry and stem cell research.
In Britain she met her future husband, a biologist from Western Australia and they have two children. The family returned to Perth in 1996.
Den Hollander succeeded Professor Sally Walker on 19 July 2010.
